Custom production of recombinant viral vectors with your sequence

The vector core takes care of your vectors production from recombinant viral genome construction to purified final product. Our production and purification processes for recombinant AAV vectors (based on viral particles separation on cesium chloride gradients) are regularly optimized, in particular for large-scale productions intended to use in large animal studies.

AAV-based viral vectors
The vector core can produce recombinant AAV with capsid from serotypes
  • 1 (AAV2/1),
  • 2 (AAV2/2),
  • 3 (AAV2/3B),
  • 4 (AAV2/4),
  • 5 (AAV2/5 and AAV5/5),
  • 6 (AAV2/6),
  • 7 (AAV2/7),
  • 8 (AAV2/8and AAV2/2i8),
  • 9 (AAV9) and
  • 10 (AAVrh2/10)
  • AAV2/DJ
  • AAV2/Anc80
  • and all vectors needed on request.

It is possible to produce ssAAV (single-stranded) and scAAV (self-complementary, i.e. double-stranded) vectors for all these serotypes.

Human adenovirus type 5 (Ad5)-based vectors

We produce first generation (ΔE1/ΔE3) adenoviral vectors containing your sequence inserted in place of the E1 region. 

Primary cells immortalization and transduction

Cell immortalization is achieved through serial infections of the cells with an MLV-based retroviral vector expressing SV40 T antigen and neomycin resistance. Stable transduction of your cells with your gene of interest can be achieved by recombinant lentiviral vector infection. A cell bank is then generated from either the selected cell population or a single cell clone.

Provision of biological materials

  • Recombinant viral vectors samples: we provide standard quantities of recombinant AAV and Ad5 vectors expressing GFP, Cre or LacZ reporter genes. We also have ΔE1/ΔE3 Ad5 control virus (without transgene) and wild-type Ad5 virus.
  • Cell lines: HEK293, HeLa, HeLaRC32, …
  • Plasmids

Technical and scientific advice for the design and optimal use of the viral vectors

Costs depend on your Institute (academic, private).

To order vectors

  1. Send your request by email to : ltg.production@univ-nantes.fr - Françoise Balter
  2. You will receive a technical proposal to fill and get it back signed after validation by the head of the production team
  3. After reading your form, we will contact you if we need more information and then, you will receive the quote to get it back signed.
  4. Your order will integrate our planning of production, when we will receive :
    - the purchase order,
    - the technical proposal signed,
    - the material (plasmid, DNA, aliquot of virus)
    Then we will give you a timing.
  5. During production, you will receive news about your vectors regularly
  6. Finally, when the vector will be ready to be sent, a date for invoice will be proposed and the shipment organized by ourselves (as your prefer)
  7. When you will receive the samples, you will also receive :
    • A purchase order from Capacité SAS (University of Nantes)
    • A second purchase order from the transporter

We archive all the production dossiers, necessary if you need informations on old productions.
